How does Colchicine work to treat acute gout?

In gout, colchicine concentrates in neutrophils (PMNs) and inhibits microtubule formation. This prevents neutrophils from migrating to inflamed joints and exerting their activity there. Neutrophils that migrate to the joint cause inflammation and since gout isn’t an infectious process, you want to stop neutrophils from going there and causing the inflammation, which is really the cause of gout symptoms.
